Instructions for Using the Genesys 10UV Spectrophotometer

 

Accomplished by Instructor or T.A.

1.      Turn on the spectrophotometer.  The power switch is located on the back of the machine, lower left side.  The spectrophotometer will automatically perform some diagnostics and will need to warm up for 30 min before measurements can be made.

2.      When finished, shut off the machine using the same power switch on the back.

 

Measuring Absorbance

1.      Select the wavelength by pressing the green button below Set nm on the LCD screen, entering the wavelength, and pressing the Set nm button again.

2.      Insert your cuvette into one of the cells, making sure that the optically clear side of the cuvette will receive light from the spectrophotometer’s lamp and that the cuvette is sitting on the bottom of the cell.  The optically clear side of the cuvette is the one marked with an arrow.  The metal strip should be on the left corner of the cuvette side with the arrow.

3.      Close the sample chamber.

4.      Read the absorbance of your sample by pressing the button which corresponds to the cell which holds your sample (either B, 1, 2, 3, 4, or 5).

5.      Remove your sample and replace it with a new sample and record the absorbance.

 

Note:  When the spectrophotometer is turned on, it should automatically be set to absorbance mode.  If it is not, press the green button under Change Mode on the LCD screen to select absorbance.  The type of mode is shown in the upper left-hand corner.

 

 

 

Instructions for Using the Genesys 20 Spectrophotometer

1.      Turn on the spectrophotometer.  The power switch is located on the back of the machine, lower left side.  The spectrophotometer will automatically perform some diagnostics and will need to warm up for 30 min before measurements can be made.

2.      When finished, shut off the machine using the same power switch on the back.

 

Measuring Absorbance

1.      Select the wavelength by pressing either the nm ­ or nm ¯ button.

2.      Insert your cuvette into the cell, making sure that the cuvette is sitting on the bottom of the cell and that the arrow on the side of the cuvette is facing you.

3.      Close the sample chamber

4.      Read your absorbance.

5.      Remove your sample and replace it with a new sample and record the absorbance.

 

Note:  When the spectrophotometer is turned on, it should automatically be set to absorbance (A) mode.  If it is not, press the A/T/C button to select absorbance (A).
